Particle Counting -- Counting Too Many/Counting Undesired Objects

Hello,

I have been using Image Pro Plus to count DAB stained positive cells.  I am using the thresholding tool to isolate the desired cells.  However, the software is picking up spots that are not the cells I want, as well as counting one cell as several signals.  For example: in one crypt the software is recognizing 200 positive cells (when by eye I count 11).  Is there a way I can fix this?

    Please provide a set of EXAMPLE IMAGES so that we can assist you.

    The BEST EXAMPLE IMAGES would include an ORIGINAL IMAGE (with no overlays or markings) and an ANALYZED IMAGE that shows what your goal is.
