Good afternoon

  • Hello,

Any of the file formats supported by Image-Pro Premier will work, TIFF would be the most common.

  • As Pierre said, any image file that Image-Pro can open can be analysed using any of the tools available in Image-Pro. However, you should bear in mind that file formats that use compression, such as JPEG, modify the original image data when the compression is applied and so for the sake of accuracy and data integrity you should avoid these file types if at all possible. Note also that JPEG images only support 8bit of grayscale resolution, so if you're acquiring your images in 12bit, you'll be losing some of that sensitivity when compressing the data to 8bit. Also, JPEG does not include much of the metadata that would enable you to compare results between various images, like exposure time, gain, sensor used etc.

Wherever possible, especially in fluorescence, it is best to use as close to the raw data as possible. This means that if you're using a monochrome camera to acquire the images it may be preferable to work on each channel individually based on a monochrome TIFF as captured by the camera, than to work on a colour composite where some of the ability to discriminate between the channels may have been lost.
