Intensity Calibration

Hi Guys,
I need to measure the DAB reaction intensity on an IHC color image (RGB 24bit).
First I created an intensity calibration using the Optical Density option.
I applied it to the color image and I drawn some measurement areas over the DAB zones.
I got the related measurements (Intensity meas & intensity std).
No problems.

After that I just tried to extract the Intensity channel (HSI method) of the color image.
I applied the same Intensity calibration used before. 
Using the FM I copied the same measurement areas over the mono image.
I was expected to get the same numbers but it was not.

I also extracted the 3 RGB channels creating manually the intensity channels by average (3 channels sum / 3)
The numbers are the same obtained using the HLI intensity channel but still different from the direct RGB measurement.

could anyone explain me these differences?

Which is the best solution for my application, get numbers from the original color image or from the intensity channel?

  • Hi Maurizio,

    We have a specialized app that extracts a stain from the image, I've attached it. Use this app to extract the stain and then use intensity measurements to measure optical density of that stain (note that this app works only in 9.2 version of Premier).
