Intensity/IOD measurements--"Sample?"

Hi everyone,

I was doing quantitative analysis using Integrated Optical Density for some Immunohistochemistry slides, and I'm not sure what "Sample" is calculating....I don't think it's number of cells because a lot of the cells were clumped together and unclear for counting. If anyone has experience, I would really appreciate the help. I'm using Image Pro Plus 7.0.

Answers

  • "Samples" reports the number of separate objects. If your cells are clumped together, they are reported as 1 objects. You should use different threshold, some pre-processing or separation tools (manual, watershed,...) if you want to measure every cell separately.
