Spatial calibration for images from slide scanner
I have obtained images via a slide scanner (Mikroscan) and save the files as Leica format (scn). The open correctly as VLI. Area measurements from ROI's that I extract are way off. I tried both:
1.) Scanning and extracting and ROI (from the scn file) from the ImagePro calibration slide and using autocalibration. That seems to work fine but measurement values do not change, even when I make sure to load the calibration file that I have saved.
2.) Doing autocalibration on the tif image that opens when I open the scn file, but the autocalibration says it cannot find the micrometer.
Is there a preferred way to get a calibration from a slide scanner image?

The step 1 is correct, it will create the proper calibration for you scans. Then you have to Apply this calibration to other slide images (just loading the calibration file is not enough).
The calibration can be applied by selecting the proper calibration and clicking the Apply button in the Spatial Calibration dialog.
