counting cells DAB staining
I'm counting pHH3 in mouse colon using DAB-staining and would like some tips on how to get the program to find my cells. I've been trying using smart segmentation, but no matter what I do I get multiple positive counts even if it's only one cell. Does anyone know how to fix this? Thanks!

If your goal is to detect small objects with high concentration of DAB stain, then you can use smart segmentation based on YIQ channels. If the image is noisy, you can also create custom channels that use LowPass over YIQ. I've attached the options file with recipe that produce this segmentation:
